Ethanol-induced alterations in pancreatic blood flow in conscious dogs.

چکیده
Because ethanol abusers are susceptible to pancreatitis in animal models, and interruption of the pancreatic circulatory supply may also cause pancreatitis, we studied the effect of ethanol infusion on pancreatic blood flow. We administered ethanol 0.5 g/kg and 1.5 g/kg iv, to 35 conscious dogs (ethanol blood levels, 117 ± 10 (SD) and 297 ± 67 mg/100 ml, respectively). Measurements were made of aortic pressure and regional blood flow by the radioactive microsphere method in the pancreas, stomach, spleen, intestines, adrenal glands, and kidneys. Pancreatic flows were 172 ml/min per 100 g control, 156 ml/min per 100 g after 0.5 g/kg ethanol and 108 ml/min per 100 g after 1.5 g/kg ethanol. After the higher dose, the decrease in pancreatic flow of 64 ml/min per 100 g or 37% was statistically significant (P< 0.01); pancreatic vascular resistance rose by 113% (P< 0.001). There were no significant changes in flow or resistance in the other organs studied. Similar changes in pancreatic flow and resistance occurred after 1.5 g/kg ethanol in groups of dogs pretreated with phenoxybenzamine, an a-adrenergic antagonist; propranolol. a /J-adrenergic antagonist; cimetidine, a histamine H2 receptor antagonist; and meclofenamate, a prostaglandin synthetase inhibitor. The changes in pancreatic blood flow and resistance were substantially reversed by 25% mannitol, 3 ml/kg, iv, after infusion of ethanol. We conclude that ethanol, in high doses relevant clinically, selectively elevates pancreatic vascular resistance, probably by inducing perivascular cellular swelling. Circ Res 50: 250-256, 1982

Role of cholecystokinin in the intestinal phase of pancreatic circulation in dogs.
The regulatory mechanisms of postprandial pancreatic hyperemia are not well characterized. The aim of this study is to clarify the role of cholecystokinin (CCK) in the intestinal phase of pancreatic circulation. Pancreatic, gastric, and intestinal blood flows were measured by ultrasound transit-time blood flowmeters in five conscious dogs. Pancreatic and gastric secretion and blood pressure wer...
متن کاملAcute renovascular hypertension in conscious dogs. Interaction of the renin-angiotensin system and sympathetic nervous system in systemic hemodynamics and regional blood flow responses.
The effects of acute renovascular hypertension on the sympathetic nervous system, regional blood flow and cardiac function were studied in conscious dogs submitted to renal artery occlusion by inflation of a cuff implanted previously around one renal artery. We then compared the alterations in plasma renin and catecholamine levels and in the various hemodynamic parameters induced by those maneu...

The effects of intravenous ouabain (G-strophanthin), 20 /ng/kg, on the superior mesenteric, renal, and external iliac beds were studied over a 30minute period in eight normal conscious dogs before, and in five of them after, cholinergic blockade and in six conscious dogs with heart failure produced by tricuspid avulsion and progressive pulmonary stenosis. Blood flows were measured with Doppler ...
متن کاملPancreatic islet blood flow in conscious rats during hyperglycemia and hypoglycemia.
Anesthesia affects general hemodynamics and regulation of organ perfusion. We used colored microspheres to measure pancreatic islet blood flow in conscious rats at two time points, during either hyperglycemia or hypoglycemia. This method, using black and green microspheres, was validated by comparison with previous microsphere experiments and by lack of effect of a nonmetabolizable glucose anal...
